Cefacetrile is primarily used as an antibiotic for treating various bacterial infections in humans and animals. Its applications include:

• Treatment of skin infections
• Management of respiratory tract infections
• Use in veterinary medicine for mastitis in dairy cows.

Despite its effectiveness, cefacetrile has seen reduced use due to the availability of newer antibiotics with improved efficacy and safety profiles.

The biological activity of cefacetrile is primarily attributed to its ability to bind to penicillin-binding proteins (PBPs) on the inner membrane of bacterial cell walls. This binding inhibits the synthesis of peptidoglycan, a critical component of bacterial cell walls, leading to cell lysis and death. Cefacetrile exhibits a peak plasma concentration of approximately 15 mg/L following an intramuscular dose of 1 g, with about 25% protein binding in plasma. Its elimination half-life is around 1.2 hours, and approximately 72% of the drug is excreted renally.
