The biological activity of 2,4,6-trimethylaniline raises concerns regarding its toxicity. It has been identified as moderately toxic when ingested, with reported oral LD₅₀ values of approximately 590 mg/kg in mice and 743 mg/kg in rats. Additionally, it can cause skin and eye irritation and has been associated with methemoglobinemia—a condition that affects the blood's ability to carry oxygen. Long-term exposure may lead to cancerous effects; studies have shown that it can induce liver tumors in laboratory animals. Furthermore, it has demonstrated weak mutagenic properties in certain assays involving bacterial strains.
